TransLvTM Lentivirus qPCR Titration Kit提供了一種利用qPCR方法檢測慢病毒轉(zhuǎn)導(dǎo)細(xì)胞中整合的前病毒序列拷貝數(shù)的方法。該方法簡單快速、靈敏高效，兼容第二代和第三代HIV-1慢病毒包裝載體。慢病毒包裝后轉(zhuǎn)導(dǎo)易感細(xì)胞 (如HT1080，HEK-293T等)，提取慢病毒轉(zhuǎn)導(dǎo)細(xì)胞基因組，進(jìn)行qPCR反應(yīng)，通過標(biāo)準(zhǔn)曲線Ct值計(jì)算整合的前病毒拷貝數(shù)，進(jìn)而得到確定的慢病毒滴度。
以梯度稀釋的標(biāo)準(zhǔn)品為模板進(jìn)行擴(kuò)增得到標(biāo)準(zhǔn)曲線，結(jié)果顯示，TransGen產(chǎn)品擴(kuò)增效率高，線性關(guān)系好。
